根据官方先前介绍所述，本作《JOJO 的奇妙冒险：全明星大乱斗 R》将提供来自原作登场的 50 位角色，玩家能够体验到每个故事中的热门战斗，并且看到来自不同宇宙的角色们互动。而在游戏模式方面，分别包括了全明星模式、街机模式、线上模式、对战模式、练习模式以及画廊模式等等，整体来说本作会提供非常丰富的游戏内容，让玩家有着与众不同的格斗对战体验。
